PACAP27 and PACAP38 have been shown to have neurotrophic effects on several neurons and possibly also in the CNS. PACAP is known to exert its biological effects by interacting with its specific receptors. The PACAP type-1 receptor is coupled to a phospholipase C and thus leads to intracellular calcium mobilization. The PACAP type-2 receptor is coupled to a mitogen-activated protein kinase, thereby leading to the activation of MAP kinases. We therefore investigated the influence of PACAP on the number of trigeminal ganglion neurons. Cell counts were performed using a stereological method. PACAP27 significantly increased the number of neurons in the ganglion at the concentrations used (100 nM-1 microM), with maximal effects at 300 nM. PACAP38, on the other hand, had no effect at concentrations up to 100 microM. PACAP38 at the concentrations used did not affect the survival of the neurons. Maximal effects of PACAP27 were obtained after 10 days in culture. These data indicate that PACAP38 and PACAP27 have distinct actions on different types of neurons.The B.C. B.C. Supreme Court Justice Kenneth Affleck ruled the proposed new electoral system is “repugnant” to the province’s charter, violating sections of the Charter of Rights and Freedoms.
Affleck, who refused to overturn the previous June’s vote that had approved the proposed changes, said the legislation does not “meet the threshold test” in terms of leaving the Liberals only one option on the ballot — no more.
